Tranexamic acid is a new generation molecule that inhibits melanin synthesis by interfering with the interaction between melanocytes and keratinocytes through inhibition of the plasminogen/plasmin system. External skin disorders, such as ultraviolet radiation or injuries (e.g., acne), cause the keratinocytes in our skin to produce plasminogen, which initiates several processes such as inflammation, increased tyrosinase activity, melanocyte differentiation, and the transport of melanosomes to the upper skin layers. Tranexamic acid slows these processes by interfering with plasmin activity, thus indirectly inhibiting melanin formation. Studies have shown that tranexamic acid, when applied topically, exhibits results similar to hydroquinone in reducing hyperpigmentation. It is also one of the most proven ingredients for the treatment of melasma. Hydroxyphenoxypropionic acid (Radianskin) complements tranexamic acid because it has an innovative mechanism that inhibits the release of melanin by melanocytes and is an alternative to tyrosinase inhibition. HPA reduces melanin formation by up to 86% and shows a greater pigmentation effect than ascorbyl glucoside, kojic acid, and arbutin, and efficacy equivalent to hydroquinone without the cytotoxicity. The formula also contains exfoliating acids such as mandelic acid (3%) and salicylic acid (0.3%) that gently remove dead cells from the upper layer of the skin, enhancing the performance of tranexamic acid and hydroxyphenoxypropionic acid, thus accelerating the reduction of hyperpigmentation and melasma.
